ABSTRACT

Objective:To explore the neuroprotective effect and mechanism of Buyang Huanwu Tang (BYHWT) on experimental autoimmune encephalomyelitis (EAE) at different stages. Method:The 36 female C57BL/6 mice were immunized subcutaneously with myelin oligodendrocyte glycoprotein peptides (MOG35-55),then randomly divided into 9, 17, 28 d EAE control group. Each BYHWT group was orally given drugs on the 3rd day after immunization (50 g·kg-1·d-1), and EAE control group was given the same volume of normal saline in the same way once a day for 9, 17 and 28 d after immunization. The effect of BYHWT on EAE mice was observed with internationally accepted clinical score. Brain and spinal cord specimens were collected at 9, 17 and 28 d after immunization. The neuroprotective effect of BYHWT was observed by hematoxylin-eosin(HE)staining and solid blue staining (LFB). The expressions of BDNF and GAP-43 in spinal cord and brain were detected by Western blot. Result:After treatment, BYHWT can significantly inhibit myelitis cell infiltration and alleviate myelin loss. Compared with EAE group, the expression of Nogo-A in the spinal cord of each BYHWT group was significantly down-regulated (PPPPConclusion:BYHWT can improve the local nerve growth microenvironment and promote the expression of NTFs, reduce the expressions of neuroinhibitory factors, and play a role in neuroprotection.

ABSTRACT

Objective To investigate the need, methods and effects training on perioperative point-of-care tran-sthoracic echocardiography (POC-TTE) in anesthesiologists.Methods In 2013, forty anesthetic attendings of Pecking Union Medical College Hospital (PUMCH) had two rounds of transthoracic echocardiography(TTE) training. In the following year, six anesthetic attendings from PUMCH experienced in intraoperative transesopha-geal (TEE) were trained by cardiologists as TTE instructors. In 2015, we conducted a survey about basic TTE education prevalence and need. Based on the results of the survey, POC-TTE workshops were hosted from 2015 to 2017. The didactic lecture, simulation practice, as well as hypotension differential diagnosis were included in those workshops. Different teaching methods were tested for their effectiveness. Results A total of 95 surveys were collected. Fifteen (15.8%) participants reported their hospital had the resources for perioperative POC-TTE exams; fourteen(14.7%)reported basic knowledge about ultrasound or other images techniques. Eighty (84.2%) acknowledged a lack of understanding about perioperative TTE. Four rounds of POC-TTE work-shops were hosted by the anesthesia department of PUMCH and a total of 83 trainees participated. Sixty three participants finished all training and testing sessions. Forty eight percent of the participants voiced the need for more hand-practice opportunities, 39% suggested that training covering more basic ultrasound knowledge, 22.5% suggested more frequent training and testing. Conclusions These training methods may increas the prevalence and acceptance of perioperative POC-TTE application. It's learning outcome has positive impact on patients' prognosis.

ABSTRACT

Aim To investigate the expression of formyl peptide receptor-2 (FPR2) in lipopolysaccharide (LPS)-induced-BV-2 cells,and detect FPR2's influence on inflammatory response induced by LPS.Methods After 1 mg · L-1 LPS acting on BV-2 cells at 12 h,the extrinsic inflammatory model was established.We used the Western blot assay to test the levels of FPR2 protein.And the expressions of phosphorylated NF-κB,TNF-α and IL-1β were investigated when the LPS-induced-BV-2 was incubated with FPR2's agonist MMK-1 and antagonist Boc-2.Transwell assay was also used to detect the LPS-inducedBV-2 migration induced by MMK-1 and Boc-2.Resuits LPS up-regulated the expression of FPR2,and when its agonist was acted on LPS-induced-BV-2,the levels of phosphorylated NF-κB,TNF-α and IL-1β were significantly higher than those of LPS group.In addition,the chemotaxis of LPS-induced-BV-2 also increased by MMK-1.These effects were abolished by Boc-2.Conclusions LPS can increase the expression of FPR2 on BV-2 cells,and FPR2 enhances the inflammatory response induced by LPS.

ABSTRACT

AIM To establish an HPLC method for the simultaneous content determination of echinacoside,verbascoside,aucubin,pinoresinol diglucoside,epimedin A,epimedin B,epimedin C,icariin and baohuoside Ⅰ in Jingang Tablets (Cistanches Herba,Eucommiae Cortex,Epimedii Folium,etc.).METHODS The analysis of 50% methanol extract of this drug was performed on a 30 ℃ thermostatic Agilent TC-C18 column (250 mm × 4.6 mm,5 μm),with the mobile phase comprising of (methanol-acetonitrile)-0.2% phosphoric acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelengths were set at 330,208 and 270 nm.RESULTS Nine constituents showed good linear relationships within their own ranges (r > 0.999 0),whose average recoveries were 96.64%-100.08% with the RSDs of 0.71%-1.76%.CONCLUSION This sensitive and accurate method can be used for the quality control of Jingang Tablets.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expression of apoptosis-related protein Bcl-w in adenocarcinoma of the small intestine, and the apoptotic effect of Bcl-w siRNA on small intestinal adenocarcinoma cells HuTu-80.</p><p><b>METHODS</b>Forty-two tissue samples were examined in our study, including 7 cases from human small intestinal adenocarcinoma, and 35 cases from normal small intestine served as control. The expression of Bcl-w was detected by immunohistochemistry (IHC). Western blot analysis was performed to confirm whether Bcl-w siRNA could effectively down-regulate Bcl-w protein after HuTu-80 cells were transfected with Bcl-w siRNA. The cells were treated with chemotherapeutic agent 5-Fu to observe whether Bcl-w protein-silecing affects the pro-apoptotic effect of 5-Fu. Flow cytometry analysis was used for assessment of apoptotic rate of HuTu-80 cells cultured with Bcl-w siRNA alone, with 5-Fu alone, and with combination of Bcl-w siRNA and 5-Fu, using untreated HuTu-80 cells as control.</p><p><b>RESULTS</b>The positive rate of Bcl-w expression was significantly higher in small intestinal adenocarcinoma than that in normal tissue (85.7% vs. 25.7%, P=0.005). Compared with the control group, Bcl-w siRNA transfection effectively down-regulated the expression of Bcl-w protein (P<0.05). The apoptosis in HuTu-80 cells was not increased significantly in Bcl-w-/-cells compared with that of control group (12.4±2.2)% vs. (8.6±1.7)% (P>0.05). However, compared with the 5-Fu group, the apoptosis in HuTu-80 cells was effectively enhanced after combination treatment with Bcl-w siRNA and 5-Fu (45.7±2.1)% vs. (71.6±3.2)% (P<0.05).</p><p><b>CONCLUSIONS</b>Bcl-w protein plays a significant role in the carcinogenesis of human small intestinal adenocarcinoma. Down-regulation of Bcl-w protein in small intestine adenocarcinoma HuTu-80 cells leads them susceptible to 5-Fu.</p>
